Preferential expansion of primed human γδ~+T cells in culture

Aim To establish one pattern of human peripheral γδ+T cells growth in vitro. Methods Without the stimulation of specific γδ+TCR ligands, the γδ+T cells in the cultured peripheral blood mononuclear cells(PBMC) of 10 selected donors preferentially proliferated in a dose dependent manner to IL 2. After 6 14 days of culture in the medium containing 3×105U/L IL 2, the percentages of γδ+T cells rose to 38% 78%with the increase of cumulative cell numbers. Results RT PCR analysis of the expanded γδ+T cells from 3 typical donors showed that they expressed TCR Vδ2 gene fragments. This kind of γδ+T cell expansion was similar to the memory αβT cell growth in vitro, which might represent the primed state of γδ+T cells in vitro. Conclusion This phenomenon will be beneficial for the understanding of γδ+T cells positions in immune system and significance in evolution.
