DR4-DRB1 allele typing in donors and recipients of kidney-transplantation

Aim To study HLA DRB1 alleles in renal allograft which are most important in match typing. Methods A pair of primers specific for DR4 were synthesized and DR4 positive DNA samples from donors and recipients of renal allografts were amplified by PCR with the primers. Then, amplified PCR products were digested with restriction endonucleases Sac II,Ava II, Hinf I, Hac II, Hph I and Mnl I, respectively. Results In accocding to the cleaved rusults and RFLP patterns, DR4 alleles can be subdivided into eleven alleles(DR4-DRB10401～DR4 DRB10411). Conclusion This method is rapid, simple and accurate and is useful for matching type of HLA DRB1 alleles of DR4 positive donor and recipient in renal allograft.
