Extraction and Purification of TGF β and Experimental Observation of Its Bone Growth Stimulating Effect

TGF β exhibits a remarkable diversity of biological activity. Platelets are a major storage site for TGF β. The aims of this study were to isolate TGF β from bovine platelets and demonstrate its biologic activity. TGF β was isolated from bovine platelets by acid/ethanol extraction, cation exchange and gel filtration chromatography. As determined by SDS PAGE, the protein was mainly 25 kD. TGF β was injected subcutaneously in mice. The blocks of multipore ceramic bovine bone(CBB) were prepared and selected as the carrier for TGF β. The complexes of TGF β and CBB were implanted into the thigh muscle of mice. Results showed that TGF β caused formation of granulation tissue at the site of injection. TGF β could also stimulate proliferation and differentiation of mesenchymal cells within the periosteum, chondrogenesis and endochondral osteogenesis. These data demonstrate that TGF β plays an important role in soft tissue repair, bone fracture healing and bone defect repair.
