Immunogenicity of a Hybrid Plasmodium Falciparum Recombinant Antigen

Li quan zhen et al. constructed a hybrid gene(HG)which codes for several T/B cell epitopes of MSA1/1, MSA2/2, RESA and universal T cell epitopes of IL 1, tetanus toxoid. The HG was cloned into expression vector pWR450 1, and expressed in E.coli to produce a fusion protein(HRA)(fused to β galactosidase) with molecular weight of 65 kD. The humoral immunity in BALB/c and C57BL/6 mice induced by HRA and soluble merozoite antigen(SMA) were investigated. BALB/c mice produced prominent anti HRA antibody after immunized with HRA emulsified with Freund′s adjuvant or HRA alone. They could also produce antibody to SMA significantly. Inversly, BALB/c mice injected with SMA could not raise antibody to HRA. C57BL/6 mice could also raise antibody to HRA significantly after immunized with HRA. These results suggest that HRA is immunogenic and may overcome MHC restriction.
