Cloning and Sequencing of the Heavy Chain Gene Fragment of an Anti hTNF α Monoclonal Antibody

We report here the nucleotide sequence of the heavy chain gene fragment of an anti hTNF α monoclonal antibody. The gene fragments were amplified with two different pairs of primers by RT PCR, from total RNA of cultured hybridoma cell line E6 which secrets the monoclonal antibody. This heavy chain gene fragment consists of 696 bp, including 5′ untranslated sequence, leading sequence, V H gene segment, D gene segment, J H1 gene and part of Cγ1 gene sequence. By comparing with the published data in EMBL Gene Bank 1995, it was confirmed as a rearranged mouse Ig heavy chain gene fragment, and the V H region gene in it appears to be a new V region gene sequence.
