Establishment of three-dimensional culture model by HaCaT keratinocytes carrying HPV11 genome and expression of HPV 11 major capsid protein L1 in this model

Objective To study the expression of HPV 11 major capsid protein L1 in the skin equivalent model established by keratinocytes carrying HPV11 genome (HPV11.HaCaT cells) in vitro, which would serve as the basis for future studies of HPV replication system in vitro and anti-viral drugs screening model. Methods Cultured fibroblasts were mixed with rat tail type I collagen to form a dermal equivalent, HPV11.HaCaT cells were seeded on the surface of the dermal equivalent and cultured three-dimensionally. After 15 days, the skin equivalent established by HPV11.HaCaT cells was harvested to make frozen section in comparison with that established by cultured normal HaCaT cells. HE staining was performed to make histological evaluation, and immuno-histochemical staining was applied to study expression of L1 protein, pan-cytokeratin and keratin 10. Results The results of HE staining showed significant increase of keratinocytes layers, associated with a tendency of differentiation. The immunostaining for L1 capsid protein, pan-cytokeratin and keratin 10 showed positive reaction in the epidermal equivalents section of HPV11.HaCaT cells; the immunostaining for pan-cytokeratin and keratin 10 showed positive reaction while L1 capsid protein showed negative reaction in the epidermal equivalents section of normal HaCaT cells. Conclusion HPV11.HaCaT cell differentiation was induced by three-dimensional culture. Markers of cell differentiation and expression of HPV11 capsid protein L1 were found in the skin equivalent model established by HPV11.HaCaT cells.
