Percutaneous absorption of the Compound Miconazole Nitrate and Hydrocortisone Ointment in vitro

Objective To establish high performance liquid chromatography(HPLC)methods for determining the transdermal absorption of Compound Miconazole Nitrate and Hydrocortisone Ointment.Methods Miconazole nitrate and hydrocortisone were quantified by HPLC,and the HPLC method was verified.The permeation of the ointment was studied on rat skin with Franz-type diffusion cell.Percutaneous absorptionin vitro was evaluated by transdermal absorption accumulation of miconazole nitrate and hydrocortisone.Results Miconazole nitrate and hydrocortisone showed good linearity in the ranges of 0.10-12.51(r=0.999 5,n=5)and 0.20-25.12 mg·L-1(r=0.999 6,n=5),respectively.Those drug solutions were stable under room temperature on 72 h.The average recoveries of miconazole nitrate and hydrocortisone were 100.4% and 100.1%,respectively.The prescription showed no significant difference for transdermal absorption accumulation when compared with reference preparation in 36 h.Conclusion The HPLC method is feasible to study the compound micronazole nitrate and hydrocortisone transdermal absorption.
