Studies on the Redox Properties of Nitrogenase from Azotobacter vinelandii

The redox properties of nitrogenase complex and its components from Azotobacter vinelandii have been studied by using optical titration. stepwise potentiostatic electrolysis and controlled potential coulometry. It was found that Av2 has a midpoint potential (Em)=-265±22 mV, with one electron transferred. When MgATP is present, the Em is lowered to -378±20 mV and two electrons are transferred. MgADP can also lower the Em to -326 mV only one electron is transferred in the redox process. Avl shows two redox regions with Em=-36 mV and -326mV. With the presence of ATP generating system, the Avl-Av2 complex has two redox regions, Em2=-290 mV and Em2=-452 mV, corresponding to transferring 6.5 and 5.1 electrons respectively.
