Acetylene Reduction Activities of Immobilized Azotobactor vcnilandii-OP Cell

C2H4 reduction activities of Azotobacter venilandii-OP cells immobilized by several methods including entrapment within alginate gel, agar gel, gelatin and adsorption to ion exchange cellulose, have been compared. Sodium alginate entrapment has been proved to be the best. Specific activity of the alginate-immobilized cells was estimated to be 11.8 μml/h/1010 cells in our hands. C2H2 reduction activity could be demonstrated up to fifteen days. Optimal conditions for C2H2 reduction appeared to be 30℃, pH7.5, and a Km value of 20μM was obtained. Effect of some respiration inhibitors, nitrogenous salts and CO2 on C2H2 reduction activity have also been examined.
