Establishment of Optimal Reaction System of RAPD in Walnut

Two methods(CTAB and reformed CTAB)were adopted to isolate genomic DNA of Walnut(Juglans rejia L.). The results show that the reformed CTAB is more effective to remove most of polysaccharides in the cytoplasmic and to inhibit the formation of oxidized polyphenolic compounds. Based on the genomic DNA extracted from Walnut, essential factors affecting the result of RAPD assay were tested and compared. An optimal reaction system suitable for the assay and usage of RAPD in Walnut was established.
