Research on Tissue Culture and Plant Regeneration of Hypocotyl in Brassica Napus L. of Xinjiang

In our experiment, the hypocotyl of Brassica napus L. 1 khpll strain is taken as explant.Based on application of differentiation medium induction seedling of 6-BA with different concentration and NAA hormone combination,the regeneration,differentiation of 6-BA and NAA combination with different hormone and concentration were found obviously different. The seedling age of sterile seedling of seeds ,pre-cultivation before 2.4 days of induction differentiation, adding AgNO_3 etc.were systematically analyzed.The results were shown that Hypocotyl of sterile seedling was in advance cultivated for 3 days in cultivating base suitable for differention of MS_o + 3.0 mg/L 6 - BA + 0.2 mg/L NAA+3.5 mg/L AgNO_3 , the regeneration and differentiation are separately 35% and 122.5%.System of high frequency regeneration plant of Brassica napus L. 1 khpll strain was preliminarily established,which contributed good conditions to develop biological technique breeding work for good quality of Brassica napus L.
