A COMPARATIVE STUDY OF PITUITARY GH GENE EXPRESSION BETWEEN BROILER AND LAYER CHICKENS DURING EMBRYONIC AND NEONATAL DEVELOPMENT

In present study,the patterns of pituitary GH gene expression were compared between broiler and layer chickens during embryonic and neonatal development.Pituitaries were taken at embryonic days (E) 14 and 18 and neonatal days (D)1,5 and 10.The expression of GH mRNA in the pituitary was detected with Northern blot analysis,and the pituitary GH content was measured with radioimmunoassay (RIA).GH mRNA was not detectable on E14 in the pituitary of both broiler and layer chicken embryos.The GH transcript (approximately 0 8 kb)was found first on E18 in the present study and the intensity of GH mRNA increased (P0 05)stepwisely thereafter through D10 in the layer,while broiler chickens exhibited a different pattern,the GH mRNA content in pituitary was increased rapidly from E18 to D1,but plateaued throughout the rest of the experiment.The developmental changes of pituitary GH contents determined with RIA were in good agreement with the ontogeny of GH mRNA in both strains of chicken.GH expression in broilers was significantly higher than that in layers at E18(P0 05),showing a positive correlation with the embryo weight during perihatch stages between E18 and D5,but a reversed correlation was observed on D10,the layer showed higher level of GH expression than the broiler,displaying a negative relationship with the early posthatch growth rate.
