ISOLATION AND CHARACTERIZATION OF CON A BINDING GLYCOPROTEINS ON THE SURFACE OF SALMONELLA TYPHIMURIUM

Salmonella typhimurium was solubilized by 2% NP 40.The extract was absorbed to Con A Sepharose 4B column and eluted by D(+) Mannose.The obtained Con A binding glycoprotein (CBG) was subjected to SDS PAGE and showed five bands with molecular weights between 15~43 kDa.The role of CBG in the adherence of bacteria to intestinal epithelium was investigated.The CBG solution (containing 0.25 μg CBG)was injected to the ligated ileum of two day old chicks and incubated for 15 min.1×10 8 bacteria were injected into the ileum and incubated for 15 min.By scanning electron microscopy,it was found that the adherence of bacteria to the intestinal epithelium was significantly inhibited by CBG.This result suggests that the CBG might be adhesin like component on the surface of Salmonella typhimurium.
