Isolation and Identification of Corynebacterium Xerosis Isolated from Yak Liver

To investigate the formation reasons of caseous nodules in the liver of a yak,bacteria were isolated and cultured from the liver,and the 16 SrRNA and rpoBgene sequences of isolated bacteria were amplified and sequenced.The biochemical phenotypes were identified by Vitek 2automatic microbiology analyzer and drug susceptibility test was performed by K-B method.Results were as follows:A Gram-positive,rod-shaped and suspected pathogen was isolated.The amplification lengths of 16 SrRNA and rpoB were 1 483 and 434bp,while the accession number were MH000696 and MH006608,respectively.The similarity of 16 SrRNA of clinical isolate to Corynebacterium xerosis was as high as 99% and phylogenetic tree showed it lied in the same branch with Corynebacterim xerosis.The results of Vitek 2fully automatic Microbial Identification System showed that it was identified as Kocuria varians,with the probability of 93%.The bacteria was highly sensitive to tobramycin,aztreonam,cefoperazone,minocycline and other medicines.In summary,the isolated bacteria is Corynebacterium xerosis.But the gene sequence,such as rpoB,and biochemical property have a certain degree of mutations.This study provides reference for the isolation and identification in the veterinary clinic,and is the foundation of path-ogenesis research of this bacteria.
