RAPID DETECTION OF XANTHOMONAS AXONOPODIS PV. CITRI WITH DOT IMMUNOBINDING ASSAY (DIA)

The capability of rapid detection of Xanthomonas axonopodis pv. citri with Dot Immunobinding Assay (DIA) was determined on the minipore filter of nitrocellulose. Effective enhancement of antiserum specificity through cross absorption of saprophytic bacteria from citrus leave and xanthomonads with colosely relationship resulted in increasing specificity of polyclonal antibodies. Lowest limitation of detection for target pathogen was 1×10 4 CFU ml -1 , Positive reactions obtained from 128 fold dilutions of soaking solution of disease citrus leave. The filter paper background can be improved via adding 0.1% Triton X-100 in TBS buffer. All 26 isolates of X.a.pv.citri from Southern China showed positive reaction with DIA but the negative controls. Detection of X.a.pv.citri with DIA indicated that there has 31.82% average positive based on 518 samples of citrus asymptomatic materials from 38 counties, 8 provinces of Southern China.
