The Study of High-frequency Regeneration System of Variegated Alfalfa

The hypocotyl of ten varieties of alfalfa(Medicago sativa L.)are used to study the effect of differ- ent cultivation condition on embryogenic callus and embryoid induction,plant development and transplant of alfalfa.The varieties of alfalfa with high embryoid induction rate and high differentiation rate are screened.They are FeiNaEr,JinHuangHou and SaiTe—6.The embryogenic callus inducted medium con- sists of changed MS+2,4-D 2 mg/L+6-BA 0.5 mg/L+sucrose 2%.The embryoid inducted medium con- sists of SH+2,4-D 2 mg/L+6-BA 0.5 mg/L+sucrose2%+LH 300 mg/L.The embryoid germinating me- dium consists of SH+GA 30.5 mg/L+sucrosel0 g/L+LH 300 mg/L.The rooting medium consists of1/ 2MS+NAA 0.3~0.5 mg/L+sucrosel0 g/L.In the system,the rate of embryogenic callus inducted is 65%~92% from hypocotyls of variegated alfalfa.The rate of Embryoid inducted is 42.3%~83.4% from embryogenic callus.The rate of embryoid germinating is 20%~75% from embryoid.The rate of rooting is 91%.The rate of replant survival is 93%.
