Effects of Progressive Strenuous Exercise on the PI3K/Akt Signaling Pathway in Skeletal Muscle of Rats

Objective To investigate the effects of exercise on the signaling pathway of PI3K/Akt and the protein synthesis after progressive strenuous exercise. Methods Male SD rats were randomly divided into two groups: control group(n=6) and progressive strenuous exercise group(n=6).The level of mRNA and protein expression of Akt,PTEN and PI3K were detected by the methods of real-time PCR and Western blotting in soleus and gastrocnemius. Results(1)The level of Akt mRNA significantly decreased in both of the muscles after progressive strenuous exercise,and the expression of Akt in gastrocnemius significantly increased;(2)No effect of progressive strenuous exercise on PTEN mRNA and it's expression was seen;(3)The expression of PI3K in soleus significantly increased after progressive strenuous exercise,but no effect of progressive strenuous exercise on the relative expression of PI3K mRNA in both of the muscles was seen. Conclusion The decrease in Akt could be occurred independently without change in PI3K,and progressive strenuous exercise could possibly switch on the inactive pathway.It was also implied that there might be other activation pathways of Akt besides the PI3K.
