Endurance Training Retards the Formation of Atherosclerosis in ApoE-gene Knock-out Mice

Objective Based on the animal model of atherosclerosis,authors studied the effect of endurance exercise on the atherosclerotic formation.Methods Forty eight 8-week old ApoE gene knock-out(ApoE-/-)mice were fed with high fat and high cholesterol diet to establish the animal model of atherosclerosis,and then were divided into four groups randomly:14-week atherosclerosis model group(group H),14-week atherosclerosis model treadmill training group(group HE),26-week atherosclerosis model group(group HH),and 14-week atherosclerosis model plus 12-week treadmill training group(group H+E).Twenty four 8-week old C57BL/6J mice were equally divided into two control groups.Mice in groups HE and H+E ran on the treadmill 60 min per day,5 days per week,at the speed of 13 m/min.The area of atherosclerosis plaque and the pathological change of aorta in the frozen sections(stained with Oil Red O and HE,respectively)were evaluated.Results(1)A number of plaques was detected in aortic tunica intima of group HE.The ratio of plaque square to total aorta square was 41.79±6.93% in group H and 25.07±7.04% in group HE which was significantly lower than that in group H(P 0.01).The pathological change was significantly lesser in group HE than that in group H.(2)The pathological change was more serious and the ratio was 58.80±6.40% in group HH.The pathological progression was relatively slow and the ratio was 50.35±3.52% in group H+E,which was significantly different from that in group HH(P 0.05).Conclusions 14-week endurance training efficiently prevents the formation of atherosclerosis in ApoE-/-mice,and 12-week endurance training retards the progression of existing atherosclerosis in ApoE-/-mice.
