Change in Mitochondrial Respiratory Chain Function in Skeletal Muscle of Rats during the HiHiLo

Objective To observe the change in mitochondrial respiratory chain function in skeletal muscle of rats during living in high altitude-exercise in high altitude-training in low altitude(HiHiLo). Methods 48 healthy male Wistar rats were randomly and equally divided into normoxic control group(C),1-week HiHiLo group(1WH),2-week HiHiLo group(2WH),3-week HiHiLo group(3WH),4-week HiHiLo group(4WH),and5-week HiHiLo group(5WH). Except the group C,the rats in 1WH,2WH,3WH,4WH and 5WH lived and trained in hypoxic environment(simulated altitude of about 2500 m)and normoxic environment respectively for1 week,2 weeks,3 weeks,4 weeks and 5 weeks. Rats in groups C and 1WH were scarified by the end of week1,and in the other groups were scarified by the end of week 2,3,4 and 5 respectively. The blood and skeletal muscles were sampled immediately after the sacrifice. The activities of creatine kinase(CK),respiratory chain complex I(NADH-Co Q reductase,CI),complex II(succinate Co Q reductase,CII),complex III(Co Q-cytochrome C reductase,CIII),and complex IV(cytochrome oxidase,CIV)in serum or muscular mitochondria were determined. Results The activity of CK increased significantly in groups 1WH,2WH and 3WH as compared with that in group C(P 0.05,P 0.01). As compared with the group C,the activities of CI,CII,CIII and CIV in group 1WH,the activities of CI,CII and CIV activities in group 2WH decreased significantly(P 0.01,P 0.05),the activity of CI in group 3WH increased significantly(P 0.05),and the activities of CI,CII,CIII and CIV in groups 4WH and 5WH increased significantly(P 0.05,P 0.01). Conclusion In this study the mitochondrial respiratory chain function in skeletal muscle improved significantly after 3 weeks of HiHiLo and reached its optimum at the 5th week of HiHiLo.
