Influence of ranitidine on nitric oxide and endothelin in rats with acetic acid-induced chronic gastric ulcer at different intervention periods

Objective To investigate the changes and correlation between nitric oxide(NO)and endothelin(ET)in rat with acetic acid-induced chronic gastric ulcer at different intervention periods by ranitidine.Methods The rat models were induced by acetic acid.The levels of NO and ET were detected.The pathological changes of gastric ulcer at different intervention periods were observed and the correlation between NO and ET was analyzed.Results As compared with that in control group,the levels of NO decreased significantly in model group(P0.01),ho-wever,the levels of ET-1 increased obviously(P0.01).After 2-week treatment with ranitidine hydrochloride capsules,the levels of NO didn't increase significantly in model group(P0.05)and the levels of ET-1 didn't decrease obviously(P0.05),which suggested that the gastric ulcer was not healed.After 4-week treatment,the levels of NO increased significantly in model group,however,the levels of ET-1 decreased obviously(P0.05 or P0.01),which showed that the ulcer healed gradually.After 6-week treatment,the levels of NO increased significantly to normal levels in model group,however,the levels of ET-1 decreased obviously to normal levels,but there was no significant difference between model group and control group(P0.05).Conclusion In rats with acetic acid-induced ahronic gastric ulcer,the balance between NO and ET was broken.Ranitidine can induce and promote the synthesis of NO,and inhibit the production of ET by feedback way,as a result,the balance of NO and ET is rebuilt and gastric ulcer is healed.
