Expression features of ERK_1 and ERK_2 in CAl area of hippocampus of mice with vascular dementia

Objective To observe the expression characteristics of extracellular signal-regulated kinase 1,2 (ERK_1 and ERK_2)in the hippocampal neurons of mice with vascular dementia(VD),and to explore its role in the pathogenesis of VD.Methods The VD models of mice were established by 3-time ischemia/reperfusion in bilateral common carotid arteries.Sham operation group was set up as control group.The behavioral scores in both groups were recorded by step-down test and water maze test at the 29th and 30th day respectively.The expressing features of ERK_1 and ERK_2 in CAl area of hippoeampus of mice in both groups were observed by immunohisochemistry method.Results The grades of learning and memory of VD mice were obviously decreased,as compared with those of sham operation group(P0.05).The expression of ERK_1 and ERK_2 in CAl area of hippocampus in model group was significantly lower than that of sham operation group(P0.05).Conclusion The decrease of expression of ERK_1 and ERK_2 may participate in the pathngenesis of VD.It is suggested that the application of the drug that could increase the levels of ERK in hippocampus may become one of the effective drugs to treat VD.
