Gene chip analysis of differentially expressed genes in AS model of ApoE mice by blood activating drugs and blood breaking drugs

Objective Using gene chip technology on load apolipoprotein E(ApoE)gene in the knockout mice atherosclerosis model in thoracic aorta in differentially expressed genes,the expression and discuss the mechanism of different doses of drugs for promoting blood circulation(Ligusticum chuanxiong and Angelica sinensis),and blood breaking drugs(tri,Ezhu)screening.Methods The C57BL/6 mice were used as blank control group(blank group).64 ApoE gene knock mice with 5 weeks according to the random number table method,were divided into model group(model group),atorvastatin atorvastatin calcium group(atorvastatin group),low Huoxue treatment group(low activity group).low broken blood medicine group(low carbon group),high blood group(high activity group),high rupture blood drug group(high rupture group),daily were given ordinary feed to duplicate the model of atherosclerosis.After successful modeling,the blank group and the model group were administrated by volume of distilled water,other groups mice were orally administered,1 times a day,for 8 weeks,in the treatment of animals,with differences in gene chip technology to detect the mouse aortic tissue expression of genes.Results The blank group compared with model group,there were 154 differentially expressed genes,including 90 genes up-regulated and 64 genes were down regulated;the model group compared with atorvastatin group,there were 96 differentially expressed genes,including 17 genes up-regulated and 79 genes were down regulated;the model group compared with the low activity group,the expression of 214 genes the difference,of which 95 genes were upregulated and 119 genes were down regulated;the model group compared with the low carbon group,there were 134 differentially expressed genes,including 87 genes up-regulated and 47 genes down regulated;high activity group compared with the low activity group,there were 236 differentially expressed genes,including 169 genes,67 gene expression;high breaking group compared with the low carbon group,there were 184 differentially expressed genes,including 88 genes up-regulated and 96 genes down regulated.AS related genes are;compared with model group,the expression of Cxcll6,Aloxl2,VcamI and Cxcr4 in blood circulation drug group and drug group was down regulated,and the expression of Alox5 ap,Cxcll2,LCAT and Ccr5 were up regulated.Conclusion Blood activating drugs(angelica sinensis and ligusticum wallichii),and blood breaking drugs(rhizoma sparganii and curcuma zedoary) show anti atherosclerotic effect and its mechanism may be related to multiple genes occur expression level changes.
