Effect of Kuijieling Ⅳ on NF-κB,TNF-α in colon tissues of ulcerative colitis rats

Objective To observe the effect of Kuijieling Ⅳ retention enema on nuclear factor kappa B(MF-κB) expression and tumor necrosis factor alpha(TNF-α) content in colon tissues of ulcerative colitis(UC) rats induced by trinitrobenzene sulfonic acid(TNBS),discussing the possible mechanism for the treatment of UC.Methods The experiments were operated in June 2015 to August 2015,In the life science and engineering laboratory of Harbin industrial university college.Choosing 12 in 138 SD rats randomly as blank control group(BC),the rest of rats were produced UC rat model induced by the method of TNBS enema and were randomly divided into model control group(MC),Kuijieling IV high dose group(HD),medium dose group(MD),low dose group(LD),positive control group(PC).HD,MD,LD group were treated respectively with high,medium and low dose of Kuijieling Ⅳ retention enema,PC group was treated by the mesalazine retention enema,BC and MC group were given for the same volume of distilled water.Using immunohislochemical method to detect the NF-κB protein expression in colonic mucosa,ELISA method lor determining the content of TNF-α and colon of rats damage rating number etc.several aspects to observe the effect of Kuijieling Ⅳ retention enema on UC rats colon tissues induced by TNBS.Results(1) colorectal colon damage:the colon tissue of rats BC clear structure,mucosa,intestinal gland rich,closely arranged;MC group made in about 1 weeks after rat colon tissue injury is the most serious;colonic mucosa necrosis,ulcer formation,massive neutrophil infiltration,PC,glands disappear;HD,MD,LD group after administration,the rat colon damage are gradually reduced.LD,MD,HD,PC group of rats colon showed intestinal mucosal epithelial defect reduced,ulcer area significantly reduced mucosal epithelial hyperplasia around repair,partial coverage,inflammatory cells decreased,granulation tissue hyperplasia significantly.Compared with the MC group,the colon injury in HD and PC group was significandy reduced.(2) NF kappa B protein expression:BC group of rat colon tissue only a small number of cells within the cytoplasm visible yellow particles,MC group cytoplasm nucleus are visible brown yellow and brown particles.With the passage of time,the expression of NF kappa B in colon tissue of rats in each group was decreased.Compared with MC group,the expression of NF kappa B in MD,HD and PC group were significantly lower(P 0.05),and there was no significant difference between LD group and MC group(P 0.05).(3) TNF-α content:TNF-α content of colon tissue in each group were higher than that of group BC,compared with the MC group,7 d after modeling,LD,MD,HD,PC group of rat colon tissue TNF-a is increased,but started from 14 d,LD,MD,HD,PC the group had decreased,PC,HD group and MC group were significantly different,and LD,MD,HD group showed an obvious dose effect relationship.Conclusion Kuijieling Ⅳ retention enema has certain protective effect on colon of UC rats induced by TNBS and its mechanism may be related to inhibit the expression of the NF-κB and reduce the inflammatory factor content of TNF-α,consequently reducing the inflammatory stimulation for colon tissues of rats by pro-inflammatory factor,achieving the protection effect on colon tissues of UC rats.
