Protective effects of monascus on cerebral hemorrhage after intracerebral hemorrhage in rats

Objective To investigate the protective effect of monascin on cerebral hemorrhage and its optimal dose.Methods From March 2016 to October 2016 at the Shanxi Medical University neural electrophysiology laboratory selected120 male SD rats were randomly divided into sham operation group,model group,low dose monascin group,middle dose monascin group and high dose monascin group with 24 rats in each group.The rat model of cerebral hemorrhage was established by injection of Gollagenase type Ⅳ.The neurological deficit scores of Garcia were measured at 1,3,7 and 14 d after operation.Results Compared with the sham group,the scores of neurological deficits in model group,low dose group,middle dose group and high dose group were significantly decreased at 1,3,7 and 14 days(F = 181.705,F =494.229,F =226.576,F =24.726,P 0.01).The brain water content was significantly higher at 1 and3 d,respectively(F =21.781,70.036,P 0.05).The amount of iron deposition was significantly increased at 3,7 and 14 days(F = 21.426,118.122,107.773,P 0.05),and reached the peak at 7-14 days,and the number of PPAR-γ immunoreactive cells increased significandy at 1,3 and 7 days(F =3.394,F =28.622,F =14.12,P 0.05)(F =33.076,F =23.17,F =47.779,F =12.182,P 0.05).The number of SRA immunoreactive cells was significandy higher than that of the control group(F =22.283,F =511.092,F =51.811,F =31.060,P 0.05).Compared wiuS the model group,the Garcia score of the high dose group increased(P 0.05),the brain water content and the amount of iron stained particles decreased(P 0.05),and the high dose group PPAR-γ,Nrf2 and SRA The number of positive cells was significantly increased at 1,3 and 7 days(P 0.05),reached the peak at 3-7 days and decreased to the level of model group at 14 days.Conclusion High dose of monascus can increase the number of PPAR-γ,Nrf2 and SRA positive cells after cerebral hemorrhage,reduce brain edema and brain iron content after cerebral hemorrhage,and improve neurological deficits after cerebral hemorrhage.
