Study on the Effect of Polygonum Perfoliatum L on the Expression of Leptin in Hepatic Tissue of Rats with Hepatic Fibrosis

Objective To explore the effect of polygonum perfoliatum L on the expression of leptin in hepatic tissues of rats with hepatic fibrosis induceded by dimethylnitrosamine( DMN). Methods A total of72 male SPF SD rats were randomly divided into 6 groups: normal group( 12 rats),model group( 9 rats),colchicine group( 10 rats),polygonum perfoliatum L high dose group( 11 rats),polygonum perfoliatum L medium dose group( 10 rats),polygonum perfoliatum L low dose group( 10 rats),after the model of HF rat induced by DMN intervened by different concentrations of polygonum perfoliatum L,conventional flakings of liver tissue from rats of all the above groups were obtained and processed by histopathological staining,and expression of leptin was detected using immunohistochemical method. Results The staining intensity and range of leptin protein in liver tissue of the rats in the model group was significantly higher than the normal group,the differences were statistically significant between the two groups( χ2= 14. 32,P 0. 05); compared with the model group,the expression levels of polygonum perfoliatum L in the polygonum perfoliatum L high dose group and polygonum perfoliatum L medium dose group significantly decreased in the liver tissues,there were statistically significant the differences( χ2= 12. 80,15. 21,P 0. 05). Conclusion Polygonum perfoliatum L has a good curative effect of anti-hepatic-fibrosis. Its therapeutic mechanism is mostly like to be adjusting the metabolism of extracelluar matrix by decreasing the expression level of leptin.
