Inhibitory Effect of Saikosaponin D on Proliferation of Osteosarcoma 143B Cells

Objective To investigate the effects of saikosaponin D( SS-d) on the growth of osteosarcoma 143 B cells,and to explore its mechanism preliminarily. Methods The proliferation of osteosarcoma 143 B cells was detected by MTT assay at 6,12,24,48,72 h after treated with different concentrations of SS-d. The expression of human osteocalcin( OCN) was detected by ELISA, and the alkaline phosphatase( AKP) activity was detected by AKP test cassette. Results Low concentrations of SS-d could significantly inhibite the growth of osteosarcoma 143 B cells in a dose-dependent manner( P 0. 05). The AKP activity test showed that there was no significant change in AKP activity in 143 B cells treated with saikosaponin. ELISA result showed that saikosaponin could attenuate the expression of OCN. Conclusion SS-d can inhibit the proliferation of 143 B cells, and the activity is the strongest, its mechanism may be unrelated to the promotion of osteogenic differentiation.
