Effect of Glimepiride on Inhibiting the Proliferation of Human Breast Cancer Cell Line MCF-7 by Down-Regulating the Expression of miRNA-214

Objective To investigate the mechanism of glimepiride in inhibiting the proliferation of human breast cancer cell line MCF-7 by down-regulating the expression of miRNA-214. Methods The human breast cancer cell line MCF-7 was cultured in vitro and divided into the blank control group and glimepiride group with low,medium and high dose. The effects of glimepiride on the proliferation of human breast cancer cell line MCF-7 cells were measured by MTT after 48 h of exposure to different doses of glimepiride.The relative expression of miRNA-214,Bax,Bcl-2 and Caspase-3 m RNA was detected by RT-PCR. The apoptosis rate of MCF-7 cells was detected by flow cytometry. Results With the increase of glimepiride dose,the proliferation rate of MCF-7 cells was decreased,and which was 54. 22% at 20 μg/m L,the difference was statistically significant compared with 0 μg/m L of glimepiride( F = 8. 52,P 0. 05). Compared with the blank control group,the relative expression levels of miRNA-214 m RNA of MCF-7 cells in glimepiride-treated group were decreased,while the relative expression levels of Caspase-3,Bax and Bcl-2 m RNA and the ratio of Bax/Bcl-2 were significantly increased,the differences were statistically significant( F = 12. 20,20. 41,6. 31,4. 42,6. 12,P 0. 05).Compared with the blank control group,the apoptosis rates of MCF-7 cells in the glimepiride-treated group were increased,and the statistic difference was found in high and medium dose of glimepiride group( F = 8. 35,P 0. 05). Conclusion Glimepiride can inhibit the proliferation of human breast cancer cell line MCF-7 with a concentration-dependent effect. Glimepiride initiates the apoptosis program and induces the apoptosis of MCF-7 cells by down regulating the level of miRNA-214.
