Inhibition of volatile oil in Capparis spinosa on proliferation of human gastric carcinoma SGC-7901 cells

Objective To study the anti-tummor effect of the volatile oil in Capparis spinosa.MethodsMTT and SRB were used to study the anti-tummor effect of the volatile oil in vitro.The typical morphology of apoptosis induced by the volatile oil in C.spinosa was abserved by reversed fluorescent microscope.The effect of the volatile oil on cell apoptosis and different stages of cell cycles in human gastric carcinoma SGC-7901 cells was studied by flow cytometer.Results The proliferation of SGC-7901 cells was inhibited after treated with the volatile oil(10,50,100,150,and 200 μg/mL)for 72 h and the IC50 was 145.5 μg/mL,GI50 was 121.32 μg/mL,LC50 was 900.96 μg/mL,TGI was 240.55 μg/mL.After SGC-7901 cells were treated by the volatile oil for 24 h,the typical morphological changes of apoptosis were observed.the apoptotic cells were detected after treated with the volatile oil(75,150,and 300 μg/mL)for 24 h.Cell cycle was also analyzed,with the results showing that the volatile oil led S stage block.Conclusion The volatile oil has the significant inhibition on proliferation of SGC-7901 cells.Its anti-tummor mechanism could be attributed to its action of initiating the apoptosis of tumor cells.
