Research on fermentation conditions for hEGF production in recombinant Escherichia coli

The effects of fermentation conditions on hEGF expression in recombinant \%E.coli\% strain JLU were examined in shake flask scale. It was found that initial glucose concentration and temperature were two key factors which affected hEGF expression. Maximum hEGF expression was observed when initial glucose was 5.0 g/L and the temperature was 32℃. Besides, high hEGF productivity was obtained when initial pH was adjusted between 6.6 and 7.3, and medium volume was 30 mL in 250 mL flask. The hEGF productivity could be further raised by 15.5% when 5 g/L glucose was replaced by 0.5 g/L glycerol as the carbon source in the fermentation medium.
