Comparison of Different Molecular Biology Methods for Detecting of Chicken Infectious Anemia Virus

In order to understand the infection of CIAV in different chicken flocks in China and the contamination of CIAV in poultry attenuated vaccines, 11 batches of 525 clinical samples and 13 batches of 125 vaccine samples submitted by the companies in the past two years were detected using conventional PCR, nested-PCR, PCR-nucleic acid spot hybridization and real-time fluorescence quantitative PCR. The results indicated that nested-PCR could provide the best sensitivity for CIAV detected by the different four methods, 59 positive of 525 samples were determined and the positive rate was 11.2%, 6 positive of 125 vaccine samples were detected and the positive rate was 4.8%. The detection rate of PCR-nucleic acid spot hybridization and real-time fluorescence quantitative PCR assay were basically consistent with nested-PCR, but the positive rate PCR was obviously lower than that of the above three methods.
