Cloning, Sequence Analysis and Tissue Expression Profiling of CALM1 Gene in Guangxi Ma Chicken

In order to analyze the coding region sequence and expression pattern of CALM1 gene in Guangxi Ma chicken. The coding region of CALM1 gene of Guangxi Ma chicken was cloned and analyzed by bioinformatics. Real-timequantitative PCR was used to detect the relative expression of CALM1 gene in different tissues of laying hens. Theresults showed that the coding region of the CALM1 gene of Guangxi Ma chicken was 450 bp in length and encodeda total of 149 amino acids. Compared with the reference sequence, the start codon and the stop codon were slightlydifferent and didn′t affect the expression, and there were two missense mutations(histidine mutation to arginine, threoninemutation to alanine). The homology analysis showed that the the sequence homology of Guangxi Ma chicken gene andGallus gallus, Sus scrofa, Bos taurus, Heterocephalus glaber, Homo sapiens, Mus musculus, Pan troglodytes, Pongoabelii, Rattus norvegicus were 98.7%, 89.4%, 84.3%, 89.8%, 90.0%, 86.7%, 89.8%, 90.0%, 86.7%. Phylogenetic treeanalysis showed that Guangxi Ma chicken had the closest relationship with the original chicken, and had the closestrelationship with cows and mice. The expression profile of CALM1 during laying period showed that CALM1 gene washighly expressed in brain, chest muscle and spleen, and was moderately expressed in the funnel and enlargement ofthe fallopian tube, heart, leg muscle, lung and intestine, but in skin, liver, ovary and fallopian tube, the isthmus, uterus,and kidney were lowly expressed. This study layed a theoretical foundation for further study on the regulation of chickenCALM1 gene in the reproductive function of Guangxi Machicken.
