Cloning and Expression Characteristics of BnNRT1.1 Gene in Ramie

NRT1. 1( nitrate transporter 1. 1) plays a very important role in the absorption and utilization of nitrate in plants. To identify the mechanism on ramie may be helpful to improve the nitrogen use efficiency of ramie. In this study,the full-length c DNA sequence of ramie Bn NRT1. 1 gene was cloned,containing 1869 bp,open reading frame( 1776 bp) and encoding 591 amino acids. The results of bioinformatics analysis showed that the protein molecular weight of Bn NRT1. 1 is 65. 25 k D and the isoelectric point is 8. 85,which is a hydrophobic protein with 12 transmembrane domains and 15 phosphorylation sites. It has important functions in signal transduction and metabolism of nitrogen translocation. Phylogenetic analysis showed that Bn NRT1. 1 gene of ramie is homologous with Prunus avium, Pyrus x bretschneideri and Morus notabilis. Tissue specific expression characteristics analysis showed that Bn NRT1. 1 was mainly expressed in Ramie roots,but it was relatively low in stems and leaves. Nitrate induction treatment showed that the expression of Bn NRT1. 1 in Longtan dama with high nitrogen use efficiency was higher than that in Qingpi ganma with low nitrogen use efficiency and the expression reached the maximum after treatment three days in different varieties. This study provides a molecular biological basis for the absorption,translocation and regulation of nitrate in ramie in order to improve the nitrogen use efficiency of Ramie.
