Study on growth Characteristics of Aortic Smooth Muscle Cell and Cardiac Fibroblast in Spontaneously Hypertensive Rats

Aim To investigate the growth characteristics of culture aortic smooth muscle cell (ASMC) and cardiac fibroblast (CFB) in spontaneously hypertensive rats (SHR). Method 16 weeks old SHR and WKY were killed after blood pressure measurement, the tissues from heart and aorta were cultured respectively. The following items were observed (1) 3H-Proline incorporation and doubling timing (DT) in CFB; 3H-TdR incorporation and DT in ASMC. (2) The effects of angiotensin Ⅱ(AⅡ) on proliferation of ASMC and CFB from SHR and WKY in the serum free medium or 2% FCS medium. Result (1) 3H-TdR incorporation of SHR ASMC and 3H-proline incorporation of SHR CFB are obviously higher than that of WKY. The DT of SHR ASMC is obviously shorter than that of WKY (30.0±1.40 vs 47.3 ±0.25, P0.01), while the DT of CFB show no difference between SHR and WKY (26.1±1.6 vs 26.6±1.9, P0.05). (2) 3H-TdR incorporation of SHR and WKY ASMC was raised to 3.3 and 2.2 times of that of control respectively by 10 -6 mol/L AⅡ(P0.01); 3H-proline incorporation of SHR and WKY CFB was raised to 3.02 and 2.3 times of that of control respectively by 10 -6 mol/L AⅡ(P0.01). 3H-TdR incorporation of SHR ASMC and 3H-proline incorporation of SHR CFB stimulated by different concentration AⅡ were much higher than those of WKY(P0.01), and the effect is corresponded with the AⅡ concentration. AⅡ can also increase the cell number of SHR ASMC and SHR CFB. The incorporation of SHR and WKY can also be promoted while the cell number remains unaltered in serum free medium. Conclusion (1) SHR ASMC and CFB have stronger proliferative ability compared with WKY; (2) AⅡ can enhance the incorporation of ASMC and CFB. AⅡ exhibits a hypertrophic effect in serum free medium and a proliferative effect in low concentration serum. (3) AⅡ has a stronger hypertrophic and proliferative effect on SHR ASMC and CFB than on WKY ASMC and CFB.
