Preparation and verification of the DGGE Marker of probiotics

Objective To design the DGGE marker for detecting type strains and verify its authenticity.Method The DNA of Lactic acid bacteria and Bifidobacteria were amplified by using their specific or universal primer,and checked out if the straps in the DGGE profiles corresponded well with those prepared from type strains.Result The DGGE profiles indicated that there was a dominant strap in each gel corresponding with DGGE marker amplified both by their specific primers and universal primer of bacteria.Conclusion The DGGE marker we prepared could be used for indicating the existence of tested type strains,and thus provide an new appoach for monitoring species change by using DGGE in microbiological study.
