Screening of high yield gamma-aminobutyric acid producing lactic acid bacteria from 'douchi' and enzymatic properties of its glutamate decarboxylase

Objective To isolate and screen high yield gamma-aminobutyric acid(GABA) lactic acid bacteria(LAB) from Yunnan traditional fermented soybean food-'douchi' and study the characteristics of its glutamate decarboxylase(GAD) in order to provide references for GABA autofermentation and immobilized production of GAD.Method A high yield GABA-producing lactic acid bacteria was screened by high performance liquid chromatography(HPLC) and its GAD properties were studied.Result A high yield GABA-producing strain,Lactobacillus plantarum YM-4-3,was successfully obtained(the yield of GABA was 5.74 mmol/L or 0.592 g/L).And the optimal fermented conditions for GAD secreting was as follows: 96 hours at initial pH 6.0 and 35 ℃ under anaerobic condition when monosodium glutamate(MSG) concentration was 3%.The enzymatic property study on crude GAD showed that the GAD secreted by L.plantarum YM-4-3 was an acidic enzyme,and it′s activity was optimal at pH 4.0 and 40 ℃,when pyrodoxal-5'-phosphate(PLP) concentration was 200 μmol/L.ConclusionThe traditional fermented 'douchi' from Yunnan province is an potential source for screening LABs with high ability of GABA conversion.This findings will be useful for the RD of new technology for fermented 'douchi' by functional lactic acid bacteria.
