Purification of total flavonoids and study in vivo and in vitro antioxidant of lespedeza cuneata

Objective To study the extraction process of total flavonoids from the lespedeza cuneata and the antioxidant activity in vivo. Methods To extract the flavonoids in the lespedeza cuneata by ultrasonic extraction,the hydroxyl radical scavenging ability and the inhibition rate of H_2O_2-FeSO_4 free radical generation system MDA were determined. Results The optimum extraction process of total flavonoids in bespedeza cuneata was: the extraction temperature was 70℃,ethanol volume fraction was 70%,the ratio of liquid to liquid was 1: 20,the extraction time was 60 min. Flavonoids extract can effectively remove hydroxyl free radicals,and had obvious inhibition effect on the liver homogenate of mice. Conclusion The extraction rate of flavonoids is 19. 43%,which has obvious scavenging free radicals and antioxidant function in vivo,and the value of medical health care should be further developed and utilized.
