Effects of Different Cryopreservation and Dilution Methods on the Development of Mouse Morulae

Two slow freezing solutions and two vitrification solutions were used in the present study to cryopreserve Kunming mouse morulae,which were loaded in plastic straws.Embryos after slow freezing were diluted out of the straws while embryos after vitrification were diluted in the straws,and the effects of different cryopreservation and dilution method on the development of embryo were compared.After cultured in vitro,differences on the developmental rates(95.3%~95.8%,98.9%) between vitrification and slow freezing with 1.5 mol/L ethylene glycol were not significant(P 0.05).Embryos after slow freezing and vitrification with EFS30 and fresh embryos were respectively transferred to recipients.Differences between the rates of embryos that develope to live fetuses of the pregnant mice were not significant(50.8%,58.3%,54.9%;P 0.05).In conclusion,in-thawing is efficient when used on embryos after vitrification.It can be used as a technical parameter in practice.
