Association of a Mutation of β3- adrenergic - receptor Gene with Obesity and Response to Dietary Intervention of Obesity in Children

Objective To determine whether a recently identified mutation in β3 - adreneric receptor gene was related to children obesity, dyslipidemia and hyperinsulinemia, and to analyze association of this mutation with response to diet intervention of obesity. Methods 311 health children aged 8-11 years including 73 overweight and 243 non - overweight were randomly selected from 4 primary schools in Beijing. A mutation of β3 - adreneric receptor gene resulting in the replacement of tryptophan by arginine at position 64 (Trp64Arg) encoding amino acid residues was detected by using the PCR- RFLP for all of them.Fasting insulin and lipids were measured and Anthropometry was carried out.47 obesity children were randomly selected from overweight children and were separated into two groups.One was diet intrvention group (36 subjects) ; the other was control group (11 subjects) . After 3 months diet intervention, the subjects of two groups were carried out anthropometry again. Results Frequency of mutated allele was similar in overweight and normal children (0.18 and 0.17, respectively) . Moreover, adjusted for age and gender, there was not significant difference in body mess index (BMI) and the levels of fasting lipids and insulin between those with mutation and without mutation among 311 chil-dren. However, the study of diet intervention in overweight children showed that the increases of weight and BMI were had significantly lower in children without mutation than in control group (2.41 ± 0.56 vs 4.43 ± 0.70, P0.05; 0.48±0.24 vs 1.55 ± 0.35, P 0.05, respectively) , but the changes in weight and BMI in children with mutation were close to the results in the control (4.32 ± 0.69 vs 4.43 ± 0.70; 1.47 ± 0.32 vs 1.55 ± 0.35) .Conclusions The Trp64Arg mutation of β3 - abr gene may contribute to the onset of obesity and forecast the result of overweight children with diet intervertion to some extent, although the Trp64Arg mutation of β3 - adr gene may not be a major factor effecting on weight in Chinese children.
