ISOLATION AND PURIFICATION OF MITOCHONDRIAL DNA OF HYBRID RAPE 3529×ORO (F_1) AND IT'S PARENTS AND A COMPERATIVE OF SOME PROPELTIES OF THEIR MITOCHONDRIAL DNA

Gather the fresh leaves of 3529×Oro(F1)and it's parents pound to pieces.After centrifuge,mitochondrion are.obtained.SDS is used to break the mitochondrion.NaClO4,CHGl3 is used to remove proteins.After dialysis,the RNase is used to get rid of mit RNAs.To make use of a few steps,the mitDNA is obtained.By way of telescope,the mitDNA is observed to be closed circular DNA molecules.By means of the ultraviolet,spectrophotography and(tke electrophoresis the DNA.samples are proved to be pure.The Tm values of their mitDNAs are respectively; 3529,35.3℃,F1,85.0℃,Oro,86.0℃.In turns the hyperchromicities are: 23.0%,29.0%,51.0% From the Tm we know the(G+C)% of them are 39%,38%,41%.The difference and approach,between mitDNA of hybrid(F1)and their parents are expressed in their shape and properties too.For example,the plant height(cm)of 3529,hybrid(F1)and Oro are respectivelyl63.1,173.2,141.8; the effective siliquae par plant are r.espectively323.7,407.0,340.5; the seeds per siliqua are respectively 13.1,14.6,11.4; the 1000-seed weights(g)are respectively 5.2,4.5,3.6; the oil contents(%)are respectively 38,8,42.6,37.1 ; the yields(g)per plant are respectively 14.2,18.7,12.4; the yield(kg)per Mu are respectively 87,157,76 etc.Therefore the hybrid(F1)is better than their parents and more approach to it's female parent 3529.
