VEGF promotes BMSCS combined with PLGA/HA stent in the treatment of radial defect in rabbits

Objective To study the efficacy of vascular endothelial growth factor( VEGF) in promoting bone marrow stromal stem cells( BMSCs) combined with polyglycolate hydroxyapatite( PLGA/HA)scaffold in the treatment of large segmental bone defect in the radius of rabbits. Methods A total of 48 three months old healthy New Zealand rabbits with weight of 2 ~ 2. 5 kg were selected without gender restrictions. to establish bilateral radial defect model with 1. 5 cm lengh,to make PLGA/HA composite porous scaffold,to culture and passage BMSCs from New Zealand rabbits and to transfect VEGF gene with sendai virus( Se V) to BMSCs,which were used as seed cells to compound with scaffold. The rabbits were divided randomly into VEGF gene transfected to BMSCs group( group A),non-VEGF gene transfected to BMSCs group( group B),single PLGA/HA scaffold group( group C),and blank control group( group D),with12 rabbits in each group. To compare callus growth and bone connection conditions were compared by X ray at one week,four weeks and eight weeks after embedding the virus. Quantity and structure of new bone formation and degradation of materials by Micro CT. The number of vascular sections and area percentage of new bone tissues and residual materials by immunohistochemical staining were compasred. Results The tissue mineral content( TMC) and trabecular connectivity density( Conn. D.) increased over the time with group A higher than group B followed by group C and group D( all P 0. 05). The percentage of area for residual materials decreased over time with group A less than group B followed by group C and group D( all P 0. 05). Conclusion VEGF can further promot BMSCs' combination with PLGA/HA scaffold for radial defect in rabbit,and thus promote new bone formation and angiogenesis.
