Endothelin release during myocardial ischemia and reperfusion in conscious rabbits with atherosclerosis

Objective The characteristics of endothelin(ET) release and myocardial injury were investigated during acute myocardial ischemia and reperfusion in rabbits with coronary atherosclerosis.Methods Fourteen control rabbits and 12 experimental rabbits suffering from atherosclerosis induced by lipid rich food were used in the present study.The rabbits were subjected to 0.5h occlusion of left ventricular branch,then reperfusion for 6h and 48h. Plasma and tissue ET was determined by radioimmunoassay.Results Plasma ET level increased from the onset of ischemia to the end of reperfusion for 48h in experimental rabbits and was higher than that in control rabbits in the same period.Plasma ET level began to step down after 6h of reperfusion in control rabbits.Tissue ET contents in non-ischemic myocardium and non-necrotic ischemic myocardium were markedly higher in experimental rabbits than in control rabbits.Less myocardial contraction band necrosis was seen at reperfusion for 6h and more focal lysis and fragmentation of myocytes were seen at reperfusion for 48h in experimental rabbits.Conclusions It is suggested that,when the events of myocardial ischemia and reperfusion occur on the basis of coronary atherosclerosis,ET release not only increases in quantity but also may last for a longer time.
