Effect of AVP_(4-9) on uptake of glutamate and gamma aminobutyric acid in cerebral synaptosomes of aged rats

Objective To study the mechanism of AVP(4-9) in facilitating memory and learning.Methods Male Sprague-Dawly rats(3 and 27 months old) were used in this study.Synaptosomes were obtained by differential centrifugation from rats brain tissue(hippocampus,frontal and temporal cortex).Synaptosomes were preincubated with AVP(4-9) or β-Alanine(β-Ala),then incubated with 3H-Glu or 3H-GABA. Results (1)AVP(4-9) increased uptake of 3H-Glu, but decreased uptake of 3H-GABA. This effect was more significant in old rats than in young rats.Young rats expressed the effect only in hippocampus, while old rats had it in hippocampus, frontal and temporal cortex. (2)β-Ala inhibited uptake of 3H-GABA in synaptosomes of both young and old rats. In old rats, β-Ala inhibited not only uptake of 3H-GABA but also that of 3H-Glu. However, β-Ala did not alter the effect of AVP(4-9) on uptake of 3H-Glu and 3H-GABA.Conclusions The results indicate that the mechanism of AVP(4-9) in facilitating memory and learning may result from regulation of Glu and GABA in synaptic level.
