The effect of Caveolin-1 on cell cycle and apoptosis in Hs578T drug-resistant breast cancer.

Objective Toinvestigate the effects of caveolion-1 gene on growth and apoptosis in Hs578T's doxorubicin-resistant (Hs578T/Dox) breast cancer. Methods The plasmid pCI-neo-cav-1 was introduced into Hs578T/Dox by lipofectin,and then selected single clone cells highly expressing Caveolin-1 level. Transfectants overexpression of Caveolin-1 was checked by Western blot. Cell cycle and apoptosis were analysis by flow cytometry. Cells proliferation were analysis by the soft agar clonogenic assay. Result The cell line overexpressed Caveolin-1 was established successfully. Transfected cells decreased markedly in G0/G1 phase (P0 .01) and increased significantly in S and (M+G2) phases (P0.01), compared with parent Hs578T/Dox cells. overexpression of caveolin-1 enhance colony formation Conclusion Caveolin-1 suppresses apoptosis and promotes growth and proliferation in Hs578T/Dox cells.Caveolin-1 also .
