PRELIMWARY STUDY ON DETECTION OF RESIDUALLEUKEMIC CELLS CARRYING POINT MUTATION OF N-RAS ONCOGENE IN ACUTE MYELOGENOUS LEUKEMIAWITH POLYMERASE CHAIN REACTION

e developed a PCR and oligotyping assay fordetection of minimal residual leukemic cells (MRLC)exhibiting a N-ras mutation in bone marrow ofpatients with acute myelogenous leukemia (AML). Theamplified N-ras sequence was detectable when as little as10pg of peripheral blcod mononuclear cells (MNC)genomic DNA was tested by this technique. With thismethed leukemic cells harboring the N-ras pointmutation were specifically detected at a minimumconcentration of 0. 1%￣ 0. 5%. Presentation DNAsfrom 10 patients with AML were screened for mutationsin codons 12, 13 and 61 of N-ras with PCR method. 3of them (30 % ) contained point mutation were identifiedall in codon 13 of N-ras. The initial results of follow upshowed that N-ras point mutation detectable by suitableoligonucleotide probes could be taken as a clonal markerto trace the MRLC carrying the N-ras mutation.
