STUDY ON THE EFFECT OF HL-60 LEUKEMC CELLSBY γhTNF-α AND RELATIONSHIP OF CELL-DIFFERENTIATION AND PROLFERATIVE SUPPRESSIONTO CHANGE OF C-MYC ONCOGENE TRANSCRIPTION

he dot blot technique of total cellular RNA with￣(32)P nicktranslated v-myc gene as probe was used tostudy the changes in transcription level of c-myconcogene as HL-60 leukemic cells were induced todifferentiation by low dose rhTNF-α. The resultsshowed that TNF with the concentration between IU/ml to 100U/ml could inhibit the transcription of c-mycmRNA in HL-60 cells, the reduction of c-myctranscription level relied on the concentration of TNFand the duration of its induction.Then, using thecellular cultivation with low concentration of fetal calfserum, in combination with cell growth kinetics, NBTreduction and ￣3H-TdR incorporation assay, we comparedthe effect of low-concentration of FCS and TNF on thedifferentiation and proliferative supression of HL-60leukemic cells in correlation with the decrease level of c-myc mRNA. We found that the decreased level of c-myctranscription was mainly correlated with HL-60 celldifferentiation induced by rhTNF-α.
