STUDY ON THE EFFECT OF HL-60 LEUKEMC CELLSBY γhTNF-α AND RELATIONSHIP OF CELL-DIFFERENTIATION AND PROLFERATIVE SUPPRESSIONTO CHANGE OF C-MYC ONCOGENE TRANSCRIPTION

he dot blot technique of total cellular RNA with ̄(32)P nicktranslated v-myc gene as probe was used tostudy the changes in transcription level of c-myconcogene as HL-60 leukemic cells were induced todifferentiation by low dose rhTNF-α. The resultsshowed that TNF with the concentration between IU/ml to 100U/ml could inhibit the transcription of c-mycmRNA in HL-60 cells, the reduction of c-myctranscription level relied on the concentration of TNFand the duration of its induction.Then, using thecellular cultivation with low concentration of fetal calfserum, in combination with cell growth kinetics, NBTreduction and  ̄3H-TdR incorporation assay, we comparedthe effect of low-concentration of FCS and TNF on thedifferentiation and proliferative supression of HL-60leukemic cells in correlation with the decrease level of c-myc mRNA. We found that the decreased level of c-myctranscription was mainly correlated with HL-60 celldifferentiation induced by rhTNF-α.
