DETECTlON OF LEUKEMlA RESlDUAL BCR/ABL mRNAWlTH NESTED POLYWRASE CHAlN REACTlON

e detected bcr/abl fusion gene with reverse tran-scription-nested polymerase chain reaction(nested PCR)assay in 33 patients with chronic myeloid leukemia(CML) in different clinical stages and 18 patients withacute lymphoblastic leukemia(ALL).Thirty one out of33 patients with CML were positive.Among 31 positivecases,21 patients showed bcr exon 3/abl exon 2 mR-NA,7 patients showed ber exon 2/abl exon 2 mRNA ,and two types of mRNA junction were detected in 2 pa-tients.Six out of 18 patients with ALL were positive.4patients showed ber exon 3/abl exon 2 mRNA,2 pa-tients showed both types of mRNA junction. The sensi-tivity of this method was 10 ̄(-6)~10 ̄(-7). With this sensi-tive nested PCR assay,we could detect the minimalresidual disease at a very early stage.
