PURIFICATION OF MULTIPOTENTIAL HEMOPOIENTIAL HEMOPOIETICSTEM CELLS WITH ANTI-FHCA-1 MONOCLONAL ANTIBODY

phthalmology Hospital of Puyang,Henan,457000 In this experiment, we detected theexpression of FH-CA-1 on mouse low density bone marrow cells(LDBM-Cs)and divided LDBMCs into three subsets according toFHCA-1 expression and wheat germ agglutinin(WGA )affinity:FHCA-1 ̄(high) WGA ̄-,FHCA-1 ̄(low) WGA ̄+ andFHCA-1 ̄- WGA ̄+ cells .After treatment with anti-FH-CA-1 monoclonal antibody(McAb)plus complement,FHCA-1 ̄(high) WGA ̄- cells were depleted from LDBMCs.Based on this characteristics,we purified LDBMC Lin ̄-FHCA-1 ̄- cells from mouse bone marrow. About 90%LDBMC Lin ̄- FHCA-1 ̄- cells obtained were WGA ̄+.Atday 12 the CFU-S yields of LDBMC Lin ̄- FHCA-1 ̄-cells was 95 times as muchas that of whole bone marrowcells.These results indicate that LDBMC Lin ̄- FHCA-1 ̄- cells are highly purified multipotential hemopoieticstem cells.
