Study on Agrobacterium-Mediated Transformation of Maize Callus

An optimized transformation system was developed for maize elite inbred lines using Agrobacterium tumefaciens mediated gene transfer by studying various important factors which affect transformation efficiency.The system was performed by transient expression of GUS gene after embryogenic callus were placed on cocultivation medium for 3 days.The study with the embryogenic callus derived from immature embryos of maize inbred line Qi 319 and R 18-599 showed that the quality of the callus played an important role in the transformation process,the callus subcultured two times and pre-cultured for 7 days were the best for transformation.In addition,the study showed that the combination of EHA 105 and Qi 319 was better than EHA 105 and R 18-599 combination for transformation,which was concluded from the detection of transient GUS expression,since the average frequency of transient GUS expression of EHA 105-Qi 319(mean=31.20%) was significantly higher than that of EHA 105-R 18-599(mean= 23.74%) based on the test(t0.05).
