FISH Karyotype Analysis of Chromosomes in Triticum aestivum L.(Guizimai No.1)

FISH(Fluorescence in situ hybridization)technique was applied to analyze FISH patterns of chromosomes in Triticum aestivum L.(Guizimai No.1),which would provide reference for the application of this variety in the breeding of new wheat varieties in the present study.The results showed that Guizimai No.1included 21 pairs of chromosomes,the pAs 1red probe signals were mainly distributed on chromosomes of A and D genome,while the pSc 119.2-1green probe signals were mainly distributed on the chromosomes of B genome.According to the distribution characteristics of these two probes on Guizimai No.1chromosomes,each chromosome could be identified accurately.Generally,The differences in signal distribution of pSc 119.2-1was showed between chromosomes 2A,5A,2B,3Band 5B of Guizimai No.1and durum wheat(AABB),and there was also a signal distribution differences on chromosomes 1D,4Dand 5Dbetween Ae.Tauschii(DD),and the signal on chromosomes 2A,5A,6B,1Dand 7D were different from those of Chinese Spring(AABBDD).The results indicated that DNA repeat sequences had polymorphisms between different wheat materials.
