Cooperation of LPS and hypoxia-ischemia on newborn rats brain damage

Objective To study whether perinatal infection cooperated hypoxia-ischemia (HI) in leading to newborn rats brain damage or not Methods ①Different dosages of lipopolysaccharide(LPS)were injected into 7-day Wistar rats to find the dosage which led to subclinical infection.②7-day Wistar rats were injected with LPS 0.3 mg/kg combined with different time of hypoxia-ischemia to measure the cerebral infarct area by microtubule-associated protein-2(MAP-2)immunohistochemistry.③7-day Wistar rats were made hypoxic-ischemic brain damage(HIBD)(HI 55 min),LPS cooperated with mild HI(LPS+HI 20 min) and the control,24 rats in each group.To measure the cerebral infarct area and intercellular adhesion molecule-1(ICAM-1)positive capillary count by MAP-2 and ICAM-1 immunohistochemistry respectively.Results ①LPS 0.3 mg/kg was the dosage which could lead to subclinical infection.②HI 20 min caused mild brain damage, but LPS+HI 20 min could lead to severe brain damage.③The cerebral infarct area was pronounced both in HI 55 min group and LPS+HI 20 min group.There was no significant difference between the two groups. ICAM-1 positive capillary count increased significantly in the ipsilateral hemisphere both in HI 55 min and LPS+HI 20 min group compared with the control( P 0.05),and it was higher in LPS+HI 20 min group than in HI 55 min group( P 0.05).Conclusions ①HI 20 min could cause mild brain damage LPS+HI 20min led to severe brain damage which meant that cooperation of LPS and mild hypoxia-ischemia lead to severe brain damage.ICAM-1 played an important role in this process.②we must pay more attention to mild asphyxia newborns if they had infection or subclinical infection in clinical work.
